Abstract/Results: RESULTS:
A new method has been developed for the assessment of several factors contributing to intuitive decisions. Technically, the method monitors the pupil dilation preceding choices made in an artificial grammar task. The analysis focuses on the acquirement of implicit knowledge related to a somatic marker. Results indicate that with the new method we can unequivocally show that a somatic marker preceding a decision is a driving “force” for the final choice. Thus the basic ingredient of the “somatic marker” model is supported.

Abstract/Results: RESULTS:
OBJECTIVES:
Previous neuroimaging findings implicate an orbitofrontal cortical dysfunction in the pathogenesis of anorexia nervosa (Uher 2003). The orbitofrontal cortex is necessary for practical decision-making in humans and the Iowa Gambling task (IGT) became established as an instrument for investigation of decision making and orbitofrontal function (Bechara 2002).
METHODS:
Using the computerised IGT, we investigated the profile of decision making performance in anorexia nervosa female patients (n=29), male patients with anorexia (n=11) long term recovered participants (n=27), healthy control females (29), healthy males (N=25). Skin conductance response during the task was measured using the PSYLAB equipment.
RESULTS:
As expected, the healthy control women made progressively more advantageous choices during the task. Patients with AN failed to show this learning effect and continued making disadvantageous choices throughout the task. Those recovered form AN, showed a normal learning curve and their performance was no worse than that of healthy controls in the IGT.
CONCLUSIONS:
The impairment of decision-making in AN is replication of previous findings (Cavedini 2004). However, decision-making in women recovered form AN was not previously reported and may elucidate the role of this cognitive function in the pathogenesis and course of the illness. This pattern of results may suggest that impaired decision-making is either a state marker of AN and improvement in decision-making is part of the process of recovery or it may be that decision-making capacity is a positive prognostic factor which makes it possible to attain full recovery. To distinguish between these two distinct possibilities, longitudinal investigation is warranted.

In this paper, we examine whether affect influences higher level cognitive processes. We review research on the effect of emotion on interpretation, judgement, decision making, and reasoning. In all cases, we ask first whether there is evidence that emotion affects each of these processes, and second what mechanisms might underlie these effects. Our review highlights the fact that interpretive biases are primarily linked with anxiety, while more general mood-congruent effects may be seen in judgement. Risk perception is also affected by negative and positive affect. Research shows complex effects of emotion on decision making and reasoning, with emotion sometimes hindering normatively correct thinking and sometimes promoting it. There are also important effects of emotion on reasoning style. We discuss key differences between the effects of incidental affect (feeling states not related to the semantic contents of the cognitive task) and integral affect (where the feeling state is caused by or linked to the contents of the cognitive task). In the conclusion, we suggest that focusing on some of the constituent mechanisms involved in interpretation, judgement, decision making and reasoning provides a way to link some of the diverse findings in the field. We also highlight important areas for future research.

Abstract/Results: ABSTRACT:
Emotional–affective and cognitive dimensions of pain are less well understood than nociceptive and nocifensive components, but the forebrain is believed to play an important role. Recent evidence suggests that subcortical and cortical brain areas outside the traditional pain processing network contribute critically to emotional–affective responses and cognitive deficits related to pain. These brain areas include different nuclei of the amygdala and certain prefrontal cortical areas. Their roles in various aspects of pain will be discussed. Biomarkers of cortical dysfunction are being identified that may evolve into therapeutic targets to modulate pain experience and improve pain-related cognitive impairment. Supporting data from preclinical studies in neuropathic pain models will be presented. Neuroimaging analysis provides evidence for plastic changes in the pain processing brain network. Results of clinical studies in neuropathic pain patients suggest that neuroimaging may help determine mechanisms of altered brain functions in pain as well as monitor the effects of pharmacologic interventions to optimize treatment in individual patients. Recent progress in the analysis of higher brain functions emphasizes the concept of pain as a multidimensional experience and the need for integrative approaches to determine the full spectrum of harmful or protective neurobiological changes in pain.

Cognitive impairment in pain through amygdala-driven prefrontal cortical deactivation

Cognitive deficits such as impaired decision-making can be a consequence of persistent pain. Normal functions of the intact amygdala and prefrontal cortex are required for emotion-based decision-making that relies on the ability to assess risk, attribute value and identify advantageous strategies. We tested the hypothesis that pain-related cognitive deficits result from amygdala-driven impairment of medial prefrontal cortical (mPFC) function. To do this, we used electrophysiological single-unit recordings in vivo, patch-clamp in brain slices, and various behavioral assays to show that increased neuronal activity in the amygdala in an animal model of arthritis pain was accompanied by decreased mPFC activation and impaired decision-making. Further, pharmacologic inhibition (with a CRF1 receptor antagonist) of pain-related hyperactivity in the basolateral amygdala (BLA), but not central amygdala (CeA), reversed deactivation of mPFC pyramidal cells and improved decision-making deficits. Pain-related cortical deactivation resulted from a shift of balance between inhibitory and excitatory synaptic transmission. Direct excitatory transmission to mPFC pyramidal cells did not change in the pain model whereas polysynaptic inhibitory transmission increased. GABAergic transmission was reduced by non-NMDA receptor antagonists, suggesting synaptic inhibition was glutamate-driven. The results are consistent with a model of BLA-driven feed-forward inhibition of mPFC neurons. In contrast to the differential effects of BLA versus CeA hyperactivity on cortical-cognitive functions, both amygdala nuclei modulate emotional-affective pain behavior. Thus this study shows that the amygdala contributes not only to emotional-affective but also cognitive effects of pain. The novel amygdalo-cortical pain mechanism has important implications for our understanding of amygdala functions and amygdalo-cortical interactions.

Abstract/Results: RESULTS:
Our first goal was to determine which brain areas represent the temporal delay of reward and to find the shape of the neural discounting function. Our results indicate that temporal delay of reward is reflected in the activation of caudate nucleus and nucleus accumbens. This is the first study to identify neural correlates of discounted value free of confounds of decision-related activation.
Our second goal was to investigate brain activation during decisions between delayed outcomes, i.e. intertemporal choice. We found that activation in the caudate and the accumbens activation was driven by chosen value. We found a strong correlation in the left caudate nucleus between the steepness of the behavioral and neural discount functions. Thus, this region might underlie individual differences in intertemporal choice behavior.
Our third goal was to look for neural correlates of a behavioral framing effect we had observed in temporal discounting. However, further testing showed this effect to be an experimental artifact that arose from the fact that no real payoffs were involved in the original experiment. Psychologists studying economic decision-making have generally assumed that subjects make similar decisions both when real payoffs are involved, and when the questions are purely hypothetical. Our results challenge this assumption and underscore the importance of using real payoffs in economic decision-making experiments.

Abstract/Results: RESULTS
Using functional magnetic resonance imaging (fMRI) in healthy volunteers, we investigated the neural bases of counterintuitive moral judgements, while controlling for the content of these judgments (utilitarian versus non-utilitarian). More specifically, we investigated the relationship between the effort required to arrive at a moral judgement, as reflected by behavioural and neural responses during moral decision-making, and two personality traits, each potentially reflecting one of the two postulated pathways to counterintuitive moral judgment (cognitive effort or emotional deficit).
Counterintuitive judgements were perceived as more difficult than intuitive judgements, whereas there was no significant difference in perceived difficulty between utilitarian and deontological judgments. At the neural level, the fMRI data suggest that previously reported differences in moral judgment are in fact largely due to their intuitiveness and not to their content. Furthermore, we show that the difficulty of making counterintuitive moral judgments is reflected in activation in the rostral anterior cingulate cortex (rACC). Importantly, rACC activation during counterintuitive judgments of a specifically utilitarian character was negatively correlated with ‘psychoticism’, a trait associated with diminished affect and social awareness, but not with ‘need for cognition’, a trait reflecting preference for complex cognition.

Neuroimaging studies on moral decision-making have thus far largely focused on differences between moral judgments with opposing utilitarian (well-being maximizing) and deontological (duty-based) content. However, these studies have investigated moral dilemmas involving extreme situations, and did not control for two distinct dimensions of moral judgment: whether or not it is intuitive (immediately compelling to most people) and whether it is utilitarian or deontological in content. By contrasting dilemmas where utilitarian judgments are counterintuitive with dilemmas in which they are intuitive, we were able to use functional magnetic resonance imaging to identify the neural correlates of intuitive and counterintuitive judgments across a range of moral situations. Irrespective of content (utilitarian/deontological), counterintuitive moral judgments were associated with greater difficulty and with activation in the rostral anterior cingulate cortex, suggesting that such judgments may involve emotional conflict; intuitive judgments were linked to activation in the visual and premotor cortex. In addition, we obtained evidence that neural differences in moral judgment in such dilemmas are largely due to whether they are intuitive and not, as previously assumed, to differences between utilitarian and deontological judgments. Our findings therefore do not support theories that have generally associated utilitarian and deontological judgments with distinct neural systems.

Abstract/Results: ABSTRACT:
Impulse control - withholding short term actions to achieve longer term goals - is an important facet of goal-directed behavior. Frontal cortex has been hypothesized to exert top-down inhibition of impulsive actions, but little is known about the nature of the neural signals responsible for such control. To address these questions, we have developed a novel impulse control task in rats and begun to investigate the causal involvement and neural correlates of frontal cortical areas.
In the impulse control task, subjects interact with a waiting port and a reward port. While waiting, two tones are generated, the first at a fixed short delay (0.4 s) and the second at a longer random delay (exponentially-distributed, min. 0.7 s). Responses after the first tone garner a reward, but the amount is 3-4 times larger after the second tone. A minimum trial onset interval was set so that waiting time did not affect trial rate. Thus, to maximize reward, subjects must withhold responding to the first tone and respond to the second.
This task produces three trial types: “failure” to wait for the first tone, “impatient” responses after the first tone and “patient” responses after the second tone. The delay of the second tone was titrated to achieve 30% patient responses (approx. 2 s mean). The timing of impatient responses varied randomly from trial to trial, approximating the distribution of second tone delays.
First, to test for areas that may be causally involved in impulse control, we infused muscimol locally to inactivate medial and dorsomedial frontal cortex (mPFC and M2). Inactivation of regions within these areas indeed led to a decrease in the waiting time and a decrease in the patient trials.
Second, to characterize neural signals related to impulse control, we made single-unit recordings from mPFC and M2 neurons, testing for signals that correlated on a trial-by-trial basis with waiting time on impatient trials. Approximately 12% of neurons in dorsomedial frontal cortex (28/238 neurons) showed activity that correlated with and predicted waiting time. We observed nearly equal proportions of positive and negative correlations.
Waiting-predictive neurons showed a variety of time courses of activity. The most frequent one was a phasic signal locked to waiting-port entry, but other neurons showed ramping activity peaking at waiting-port exit. Waiting correlated activity sometimes even occurred a few seconds before waiting port entry.
These results establish a task suitable for studying impulse control, suggest the involvement of dorsomedial frontal cortex in this function, and reveal properties of single neural activity that may be responsible for top-down control of impulsive action.

A fundamental topic in neuroscience is the nature of “free will” and how it is derived from neurophysiological processes. In this study, we investigate decision-making by examining the differences in brain activity underlying free and forced behaviours using magnetoencephalographic (MEG) recordings from subjects performing button pressing tasks that require them to exercise their choice by pushing one of two buttons in response to various cuing methods. The greatest ability to discriminate (>80% classification accuracy) free and forced trials came from MEG sensors located over the primary sensory cortices specific for the modality used to cue each trial: either visual (occipital) or auditory (left temporal), and minor non-localized differences for trials that were pre-specified. These findings suggest that primary sensory areas play a crucial part of the information processing steps organizing the production of free and forced behaviours, and, contrary to the current conceptualisation, that fronto-parietal processes may not be the principal determinants of these actions.
