Among its activities the BIAL Foundation manages the BIAL Award, created in 1984, one of the most important awards in the Health field in Europe. The BIAL Award rewards both the basic and the clinical research distinguishing works of major impact in medical research. The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology. To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665). Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world. Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries. The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project. The supported projects have originated, until April 2013, in about 600 full papers, 172 out of which were published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665). Among the BIAL Foundation fellows is worth highlighting the presence of scientists from prestigious universities from the United States, United Kingdom, Australia, Russia, Germany, Japan, France, Canada, and many others. Abstract/Results:	ABSTRACT: Emotional–affective and cognitive dimensions of pain are less well understood than nociceptive and nocifensive components, but the forebrain is believed to play an important role. Recent evidence suggests that subcortical and cortical brain areas outside the traditional pain processing network contribute critically to emotional–affective responses and cognitive deficits related to pain. These brain areas include different nuclei of the amygdala and certain prefrontal cortical areas. Their roles in various aspects of pain will be discussed. Biomarkers of cortical dysfunction are being identified that may evolve into therapeutic targets to modulate pain experience and improve pain-related cognitive impairment. Supporting data from preclinical studies in neuropathic pain models will be presented. Neuroimaging analysis provides evidence for plastic changes in the pain processing brain network. Results of clinical studies in neuropathic pain patients suggest that neuroimaging may help determine mechanisms of altered brain functions in pain as well as monitor the effects of pharmacologic interventions to optimize treatment in individual patients. Recent progress in the analysis of higher brain functions emphasizes the concept of pain as a multidimensional experience and the need for integrative approaches to determine the full spectrum of harmful or protective neurobiological changes in pain.

Abstract/Results:	ABSTRACT; Cognitive deficits such as impaired decision-making can be a consequence of persistent pain. Normal functions of the intact amygdala and prefrontal cortex are required for emotion-based decision-making that relies on the ability to assess risk, attribute value and identify advantageous strategies. We tested the hypothesis that pain-related cognitive deficits result from amygdala-driven impairment of medial prefrontal cortical (mPFC) function. To do this, we used electrophysiological single-unit recordings in vivo, patch-clamp in brain slices, and various behavioral assays to show that increased neuronal activity in the amygdala in an animal model of arthritis pain was accompanied by decreased mPFC activation and impaired decision-making. Further, pharmacologic inhibition (with a CRF1 receptor antagonist) of pain-related hyperactivity in the basolateral amygdala (BLA), but not central amygdala (CeA), reversed deactivation of mPFC pyramidal cells and improved decision-making deficits. Pain-related cortical deactivation resulted from a shift of balance between inhibitory and excitatory synaptic transmission. Direct excitatory transmission to mPFC pyramidal cells did not change in the pain model whereas polysynaptic inhibitory transmission increased. GABAergic transmission was reduced by non-NMDA receptor antagonists, suggesting synaptic inhibition was glutamate-driven. The results are consistent with a model of BLA-driven feed-forward inhibition of mPFC neurons. In contrast to the differential effects of BLA versus CeA hyperactivity on cortical-cognitive functions, both amygdala nuclei modulate emotional-affective pain behavior. Thus this study shows that the amygdala contributes not only to emotional-affective but also cognitive effects of pain. The novel amygdalo-cortical pain mechanism has important implications for our understanding of amygdala functions and amygdalo-cortical interactions.

Abstract/Results:	ABSTRACT: Uncovering the roles of the orbitofrontal cortex and the amygdala in rewardbased decision-making strategies has relied heavily in studies using discrete brain lesions. However, chronic pain patients also display risk-prone behavior even in the absence of discrete brain lesions. The main objective of this project was to study how chronic pain affects brain areas critical for reward processing. For this we developed a novel Rodent Gambling Task in which naïve animals make consecutive choices between two levers that attribute rewards of different value and probability. After a prolonged training phase in which both levers give the same reward, in the probe trial we maintain one lever at 1 food pellet with 80% probability of reward - low risk lever – while the other lever gives 3 pellets but at only 30% probability – high risk lever. We found that control animals start the task with a preference for the high-risk lever, but progressively shift to the low-risk option, while animals with orbitofrontal or amigdalar lesions, and animals with chronic pain prefer the high-risk lever. Since no brain lesion is caused by the model of chronic pain, we performed an HPLC analysis and found a decrease in the levels of dopamine and serotonin at the OFC, with no significant changes in the amygdala. We extended these findings by chronically implanting multielectrodes and recording 96 neurons in awake-behaving animals performing four sessions of the RGT before and after induction of pain. We found that the neuronal instantaneous firing rate was correlated with the probability of choosing a specific lever in the majority of the recorded neurons. Moreover, we found that the fraction of risk-sensitive neurons recorded in each session predicted the overall behavioural risk-bias of the animal. This project demonstrated that chronic pain changes the reward-aversion circuitry and that these changes are crucial for setting individual levels of risk preference. Title:	Age-related differences in the neural correlates of remembering time-based intentions

Abstract/Results:	ABSTRACT: The present study used event-related potentials (ERPs) to explore the effect of age on the neural correlates of monitoring processes involved in time-based prospective memory. In both younger and older adults, the addition of a time-based prospective memory task to an ongoing task led to a sustained ERP activity broadly distributed over the scalp. Older adults, however, did not exhibit the slow wave activity observed in younger adults over prefrontal regions, which is considered to be associated with retrieval mode. This finding indicates that age-related decline in intention maintenance might be one source of the impaired prospective memory performance displayed by older adults. An 'anterior shift' in scalp distribution of the P3 was observed in older adults, and was related to lower levels of accuracy in prospective memory performance. This relationship suggests that possible factors responsible for age-related decline in prospective memory performance include the decreased efficiency of executive/frontal functions as well as the reduced amount of resources available for the prospective memory task.

Abstract/Results:	ABSTRACT: Cognitive deficits such as impaired decision-making can be a consequence of persistent pain. Normal functions of the intact amygdala and prefrontal cortex are required for emotion-based decision-making that relies on the ability to assess risk, attribute value, and identify advantageous strategies. We tested the hypothesis that pain-related cognitive deficits result from amygdala-driven impairment of medial prefrontal cortical (mPFC) function. To do this, we used electrophysiological single-unit recordings in vivo, patch clamp in brain slices, and various behavioral assays to show that increased neuronal activity in the amygdala in an animal model of arthritis pain was accompanied by decreased mPFC activation and impaired decision-making. Furthermore, pharmacologic inhibition (with a corticotropin-releasing factor 1 receptor antagonist) of pain-related hyperactivity in the basolateral amygdala (BLA), but not central amygdala (CeA), reversed deactivation of mPFC pyramidal cells and improved decision-making deficits. Pain-related cortical deactivation resulted from a shift of balance between inhibitory and excitatory synaptic transmission. Direct excitatory transmission to mPFC pyramidal cells did not change in the pain model, whereas polysynaptic inhibitory transmission increased. GABAergic transmission was reduced by non-NMDA receptor antagonists, suggesting that synaptic inhibition was glutamate driven. The results are consistent with a model of BLA-driven feedforward inhibition of mPFC neurons. In contrast to the differential effects of BLA versus CeA hyperactivity on cortical-cognitive functions, both amygdala nuclei modulate emotional-affective pain behavior. Thus, this study shows that the amygdala contributes not only to emotional-affective but also cognitive effects of pain. The novel amygdalo-cortical pain mechanism has important implications for our understanding of amygdala functions and amygdalo-cortical interactions.

Abstract/Results:	ABSTRACT: Chronic stress impairs cognitive function, namely on tasks that rely on the integrity of cortico-limbic networks. To unravel the functional impact of progressive stress in cortico-limbic networks we measured neural activity and spectral coherences between the ventral hippocampus (vHIP) and the medial prefrontal cortex (mPFC) in rats subjected to short term stress (STS) and chronic unpredictable stress (CUS). CUS exposure consistently disrupted the spectral coherence between both areas for a wide range of frequencies, whereas STS exposure failed to trigger such effect. The chronic stress-induced coherence decrease correlated inversely with the vHIP power spectrum, but not with the mPFC power spectrum, which supports the view that hippocampal dysfunction is the primary event after stress exposure. Importantly, we additionally show that the variations in vHIP-to-mPFC coherence and power spectrum in the vHIP correlated with stress-induced behavioral deficits in a spatial reference memory task. Altogether, these findings result in an innovative readout to measure, and follow, the functional events that underlie the stress-induced reference memory impairments.

Title:	Chronic stress disrupts neural coherence between cortico-limbic structures

Abstract/Results:	ABSTRACT: Chronic stress impairs cognitive function, namely on tasks that rely on the integrity of cortico-limbic networks. To unravel the functional impact of progressive stress in cortico-limbic networks we measured neural activity and spectral coherences between the ventral hippocampus (vHIP) and the medial prefrontal cortex (mPFC) in rats subjected to short term stress (STS) and chronic unpredictable stress (CUS). CUS exposure consistently disrupted the spectral coherence between both areas for a wide range of frequencies, whereas STS exposure failed to trigger such effect. The chronic stress-induced coherence decrease correlated inversely with the vHIP power spectrum, but not with the mPFC power spectrum, which supports the view that hippocampal dysfunction is the primary event after stress exposure. Importantly, we additionally show that the variations in vHIP-to-mPFC coherence and power spectrum in the vHIP correlated with stress-induced behavioral deficits in a spatial reference memory task. Altogether, these findings result in an innovative readout to measure, and follow, the functional events that underlie the stress-induced reference memory impairments.

Abstract/Results:	ABSTRACT: Astrocytes are being recognized as active elements concerning information flow in the brain, however their role in cognition is still unknown. In this study we aimed to understand the influence of astrocytic function in complex cognitive processes that depend on the medial prefrontal cortex (mPFC). The rationale consisted in analysing the cognitive performance of animals with induced glial pathology in the mPFC. This animal model was obtained by bilateral intracranial injections of L-a-aminoadipate to cause astrocyte ablation in Wistar-Han rats. Cognitive functions dependent on the mPFC, such as attention set-shifting and reversal learning of non-spatial working memory, were assessed by the attentional set-shifting task (ASST). Animals with mPFC-glial pathology showed no differences in learning how to perform the task, however they learnt faster the compound discrimination stages, when compared to the controls. Regarding the reversal learning stages, drug injected animals presented more difficulties than controls. Glial pathology seems to affect the function of neuro-glial networks, which may be translated in altered behavioural outputs.

Abstract/Results:	ABSTRACT: Interest in astroglial cells is rising due to recent findings supporting dynamic neuron–astrocyte interactions. There is increasing evidence of astrocytic dysfunction in several brain disorders such as depression, schizophrenia or bipolar disorder; importantly these pathologies are characterized by the involvement of the prefrontal cortex and by significant cognitive impairments. Here, to model astrocyte pathology, we injected animals with the astrocyte specific toxin L-a-aminoadipate (L-AA) in the medial prefrontal cortex (mPFC); a behavioral and structural characterization two and six days after the injection was performed. Behavioral data shows that the astrocyte pathology in the mPFC affects the attentional set-shifting, the working memory and the reversal learning functions. Histological analysis of brain sections of the L-AA-injected animals revealed a pronounced loss of astrocytes in the targeted region. Interestingly, analysis of neurons in the lesion sites showed a progressive neuronal loss that was accompanied with dendritic atrophy in the surviving neurons. These results suggest that the L-AA-induced astrocytic loss in the mPFC triggers subsequent neuronal damage leading to cognitive impairment in tasks depending on the integrity of this brain region. These findings are of relevance to better understand the pathophysiological mechanisms underlying disorders that involve astrocytic loss/dysfunction in the PFC.

Abstract/Results:	ABSTRACT: BACKGROUND/AIM The aim of this work was to investigate the motivational intensity model (MIM) using measures of brain activation. The MIM has been extensively explored using cardiovascular psychophysiology, however the activation of the prefrontal cortex has not been explored in this particular paradigm. The aim of this study was to investigate how cognitive demand in response to working memory load affected activation of the rostral prefrontal cortex (rPFC) under two conditions: (a) control (no reward) and (b) financial reward. It was hypothesised that the presence of reward would enhance rPFC activation but only when cognitive demand was high. METHODS 40 participants (20 males) volunteered for the mixed-design study where reward group served as a between-participants factor. Cognitive demand was varied by exposing participants to a working memory task (nback) at five levels of difficulty, e.g. 0-back, 1-back, 3-back, 5-back, 7-back. Demand was estimated to be easy (0-back, 1-back), hard (3-back, 5-back) or impossible (7-back). All participants were paid £10 for taking part in the study, but half of the participants were offered an additional monetary reward of £10 that was linked to their performance on the task. Blood oxygenation in the rPFC was captured using a functional near-infrared spectrography (fNIRS) device that covered the forehead region of the participant and monitored BA10. RESULTS The results of the study indicated that blood oxygenation in the rPFC was generally higher in the presence of reward. It was also apparent that activation of the rPFC peaked at the 3-back task for participants in the control group, whereas this peak was observed at the 5-back for participants in the reward group. CONCLUSIONS The rPFC responded to both cognitive demand and the presence of a reward. This effect was consistent with the MIM where the level of effort invested into a task is influenced by the likelihood of success and the presence of an extrinsic reward.

Abstract/Results:	ABSTRACT: In this chapter, we review studies investigating the role of sleep in emotional functions. In particular, evidence has recently accumulated to show that brain regions involved in the processing of emotional and reward-related information are activated during sleep. We suggest that such activation of emotional and reward systems during sleep underlies the reprocessing and consolidation of memories with a high affective and motivational relevance for the organism. We also propose that these mechanisms occurring during sleep promote adapted cognitive and emotional responses in the waking state, including overnight performance improvement, creativity, and sexual functions. Activation across emotional-limbic circuits during sleep also appears to promote emotional maturation and the emergence of consciousness in the developing brain.

Abstract/Results:	ABSTRACT: Neurons in higher cortical areas, such as the prefrontal cortex, are often tuned to a variety of sensory and motor variables, and are therefore said to display mixed selectivity. This complexity of single neuron responses can obscure what information these areas represent and how it is represented. In this project we developed and demonstrated advantages of a new dimensionality reduction technique, demixed principal component analysis (dPCA), that decomposes population activity into a few components. In addition to systematically capturing the majority of the variance of the data, dPCA also exposes the dependence of the neural representation on task parameters such as stimuli, decisions, or rewards. To illustrate our method we reanalyzed population data from four datasets comprising different species, different cortical areas and different experimental tasks. In each case, dPCA provides a concise way of visualizing the data that summarizes the task-dependent features of the population response in a single figure. This project also involved some additional work on dimensionality reduction of electrophysiological datasets, including data from auditory cortex and from input areas to dopaminergic system in rats.

Abstract/Results:	ABSTRACT: Neurons in higher cortical areas, such as the prefrontal cortex, are often tuned to a variety of sensory and motor variables, and are therefore said to display mixed selectivity. This complexity of single neuron responses can obscure what information these areas represent and how it is represented. Here we demonstrate the advantages of a new dimensionality reduction technique, demixed principal component analysis (dPCA), that decomposes population activity into a few components. In addition to systematically capturing the majority of the variance of the data, dPCA also exposes the dependence of the neural representation on task parameters such as stimuli, decisions, or rewards. To illustrate our method we reanalyze population data from four datasets comprising different species, different cortical areas and different experimental tasks. In each case, dPCA provides a concise way of visualizing the data that summarizes the task-dependent features of the population response in a single figure.

To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665). Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world. Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

Abstract/Results:	ABSTRACT: BACKGROUND: Negative emotional responses to the daily life stresses have cumulative effects which, in turn, impose wide-ranging negative constraints on emotional well being and neurocognitive performance (Kalueff and Nutt, 2007; Nadler et al., 2010; Charles et al., 2013). Crucial cognitive functions such as memory and problem solving, as well more short term emotional responses (e.g., anticipation of- and response to- monetary rewards or losses) are influenced by mood. The negative impact of these behavioral responses is felt at the individual level, but it also imposes major economic burden on modern healthcare systems. AIMS: Although much research has been undertaken to understand the underlying mechanisms of depressed mood and design efficient treatment pathways, comparatively little was done to characterize mood modulations that remain within the boundaries of a healthy mental functioning. METHODS: In two placebo-controlled experiment, we applied daily prefrontal transcranial Direct Current Stimulation (tDCS) at five and three points in time. RESULTS: We found reliable improvements on self-reported mood evaluation. Using a new team of experimenters, we replicated this finding in an independent double-blinded placebo-controlled experiment and showed that stimulation over a shorter period of time (3 days) is sufficient to create detectable mood improvements. CONCLUSIONS: Taken together, our data show that repeated bilateral prefrontal tDCS can reduce psychological distress in non-depressed individuals.

Abstract/Results:	ABSTRACT: Previous research has demonstrated changes in neurovascular activation of the prefrontal cortex to increased working memory load. The primary purpose of the current paper was to investigate overload of working memory capacity using functional near-infrared spectroscopy (fNIRS) within the framework of motivational intensity theory. A secondary goal was to explore the influence of the correlation-based signal improvement (CBSI) as a method for correcting the influence of systemic variables. In study one, 30 participants (15 female, mean age = 21.09 years, s.d. = 2.9 years) performed a verbal version of the n-back working memory task under four levels of demand (easy, hard, very hard, and impossible). In contrast to the raw data, CBSI-transformed fNIRS data indicated that neurovascular coupling was highest at hard demand when the task was challenging but success was possible. The second study (N??=??30; 15 female, mean age = 22.4 years, s.d. = 5.3) replicated the working memory manipulation with the addition of low versus high levels of financial reward. Analyses of CBSI-transformed levels of oxygenated (HbO) and deoxygenated (HHb) hemoglobin replicated the first study at right lateral regions of the prefrontal cortex (BA46). HHb_CBSI data were significantly reduced at impossible demand for participants receiving the higher level of financial reward. The study is the first to support predictions from the motivational intensity model using neurovascular data. In addition, the application of CBSI to fNIRS data was found to improve the sensitivity of HbO and Hbb to the independent variables.
